Q: Can the pressure switched be removed from the manifold, rotated and then replaced?

Yep, I made that mistake too. It's all spring loaded. You can't replace the bottom manifold with the correct one. It's junk if you remove the 4 bolts holding the manifold. DON'T DO IT.

Q: Can the manifold be rotated?

There are 4 ports on the switch, these can be configured any way that works with your compressor and gauges and output air. I assume that is what you meant by rotate.

Q: Will this work on: old porter cable 6hp 25 gallon model: cpl6025? to replace: z-d20645 (yes, discontinued)?

You have to compare this switch with yours. Does it mount on the bottom does yours have four holes? is there a small unloader line from the tank to your switch? Check all of these.
