Can this be used on wood panel roof ?

Yea use a heat gun though some say you dont need one but you'll get a much better adhesion if you use a heat gun it's good stuff I used when the lead flashing had wore through between my wooden conservatory and the side of my house works great no leaks

No need to melt. It has a sticky back. Just unroll, peak and stick

Can I use this to seal a downpipe where rainwater pours out at a connection when there's heavy rain?

I would say no but why dont you take the connection apart and use silicone sealant..better and cheaper
