I used it in conjuction with a second layer of drywall. It needs the second layer to spread the liquid over the surface of the first layer. Look it up on YouTube. There are a few instructional videos that explain the use rather well.

I'm adding a layer of drywall to a plaster wall. Do I need to paint primer on the plaster wall first?

No reason to in regards to the Green Glue with drywall. You do want to make sure the wall is smooth though, by sanding it down if necessary.
